    Single Cell Full Length mRNA Amplification Kit can achieve enough amount of full-length cDNA through using 1 – 500 of cells or 10 pg – 10 ng of total RNA as templates. In this kit, Oligo (dT) VN Primer is used as reverse transcription primer and template-switching activity of Sc Reverse Transcriptase is utilized to add adapter sequences at the 3′ of cDNA. Subsequently, PCR amplification is performed through the adapter sequences to obtain full-length cDNA amplification products, which effectively avoids 3′ preference and rRNA contamination in the cDNA synthesis process. The obtained full-length cDNA amplification products can be used for genetic regulation information analysis such as differential gene expression, alternative splicing, and fusion genes. Single Cell Full Length mRNA Amplification Kit is compatible with sample volumes within 2.5 μl. Generally, 2 – 20 ng of cDNA amplification products can be obtained in one reaction.

It solves the technical problem that conventional mRNA-seq cannot be used for sequencing analysis of micro-samples such as single cells due to the low RNA content. N711 uses Oligo dTPrimer as the reverse transcription primer for cDNA synthesis, uses the Discover-sc Reverse Transcriptase Template-switching activity to add a linker sequence to the 3′ end of the cDNA, and then uses the linker sequence for subsequent PCR amplification to obtain full-length cDNA amplification product. It is highlighted for the uniform transcript coverage and more gene detections, and therefore more suitable for the detection of low-frequency genes and low-concentration templates.

    VAHTS HiFi Amplification Mix for MGI is a high fidelity PCR amplification master mix. The core component of this product is VAHTS HiFi DNA Polymerase. It is a new generation high yield and high fidelity DNA polymerase adapted from Pfu DNA Polymerase. It has greatly increased sensitivity, high amplification efficiency and extensive template compatibility. The amplification error rate of VAHTS HiFi DNA Polymerase is 52-fold lower than that of Taq DNA Polymerase and 6-fold lower than that of Pfu DNA Polymerase, which ensures the fidelity. The products can be widely applied to whole genome sequencing, whole exome sequencing, large fragment copy number variation analysis, microsatellite analysis, qPCR analysis, or gene chip analysis.The Phi29 DNA polymerase used in this kit is cloned from phage and has a strong strand displacement activity. Melting and replication of the DNA chain with complex structure can be achieved. Meanwhile, Phi29 has a strong chain affinity and can achieve up to 100 kb of continuous polymerization, and the amplification product can be applied to almost all of the downstream genome analysis.
